Experimental principle
The cytoplasmic membrane is a dynamic flow structure formed by the different degrees of protein embedded in the lipid bilayer. Protein and lipid molecules are combined with oligosaccharide chains to form glycoproteins and glycolipid molecules. Glycoproteins and glycolipid molecules extend to the cell surface. The branched oligosaccharide chain forms an extracellular surface (also known as glycocalyx) on the surface of the plasma membrane. Many studies have shown that molecular recognition between cells, cell growth and differentiation, immune response and tumorigenesis are all related to extracellular (branched oligosaccharide chains).
Lectin is a type of protein that contains sugar (with the exception of a few lectins) and can specifically bind to sugar. It has the function of agglutinating cells and stimulating cell division. Lectin promotes cell agglutination mainly because it can be linked to extracellular sugar molecules and form a "bridge" between cells. The addition of a sugar molecule complementary to lectin can inhibit agglutination between cells.
Experimental reagent
1. PBS buffer: Weigh NaCl 7.2g, Na2HP04 1.48g, KH2P04 0.43g, add distilled water, dilute to l000 mL, adjust the pH to 7.2.
2. 1% heparin solution: Weigh 0.1 g of heparin sodium and dilute to 10 mL of normal saline.
Laboratory equipment
Microscope, tray balance, several slides, 5 mL syringe, 2 dropper, 2 centrifuge tubes.
Experimental Materials
1. Potato tubers.
2. 2% red blood cell suspension
Thoroughly extract 1 mL of rabbit ear vein blood (5 mL syringe infiltrated with 1% heparin solution), wash 5 times with physiological saline, 3000 r/min each time, centrifuge for 5 min, and finally use the physiological volume of red blood cells deposited by sedimentation. The saline was formulated into a 2% red blood cell suspension.
Experimental procedure
1. Weigh 2g of potato peeled tubers and cut into small pieces
2. Add 10 mL of PBS buffer and soak for 2 h (the leached crude extract contains soluble potato lectin)
3. Drop a drop of potato lectin on the slide
4. Add another drop of 2% red blood cell solution and mix thoroughly.
5. Allow to stand for 20 min
6. Observing hemagglutination under low power microscope
7. PBS solution plus 2% blood cell suspension as a control experiment, observed under a low power microscope.
