Selection and use of carbon dioxide incubators Since animal cell culture fluids generally use a carbonate pH buffer system, it is required to maintain a certain concentration of CO2 in the culture environment, so the culture of animal cells generally requires a CO2 incubator.
The CO2 incubator uses two different CO2 maintenance systems. One way is to mix CO2 and air in a certain ratio and then blow it into the incubator by using an air pump. Another way is to open the CO2 valve to charge CO2 when an automatic CO2 detection control system is detected, when the CO2 concentration is lower than the required concentration. Since the former method consumes CO2 and it is not easy to accurately control the CO2 concentration, the advanced CO2 incubator uses an automatic monitoring system to maintain the CO2 concentration.
Incubators equipped with an automatic CO2 detection and control system are required to perform regular CO2 concentration testing and correction according to the manufacturer's instructions.
The insulation of the CO2 incubator is of the water jacket type and the air jacket type. The air jacket type uses an insulation system and a surface heat dissipating component to equalize the temperature inside the incubator. However, because the external electric heating system is unevenly distributed, the temperature in the incubator may not be uniform, and the temperature difference is relatively large. The water jacket type requires the user to inject a large amount of distilled water into the space of the tank wall when the incubator is installed. Since the specific heat capacity of water is very high, the advantages of the water jacket type are to help uniform heat dissipation and avoid formation of a cold zone, and to better reduce the temperature drop in the incubator after power failure. The water jacket type has a good temperature uniformity due to the water circulation for 4 weeks. Most of the new CO2 incubators are water jacketed.
Most laboratories currently use imported cell culture incubators. Although the quality of imported products is relatively guaranteed, the price is very expensive, and the after-sales service is not very convenient.
The CO2 incubator is usually equipped with a water tray to maintain high humidity. The water in the water tray should be added regularly, and the water tray should be cleaned regularly to avoid the growth of microorganisms.
In order to reduce microbial contamination, some CO2 incubators provide sterilization. Some can be heated to a higher temperature for sterilization, while others are sterilized by circulating air through an ultraviolet chamber, so that sterilization does not require disruption of the cells.
